Chitinases from Serratia marcescens and Pseudomonas stutzeri were generous gifts from Dr Roy Fuchs, Monsanto Company, St Louis, Missouri, USA. The Sa. marcescens chitinase was prepared by cloning the gene into Escherichia coli, where it was expressed and the protein purified to yield a single 57 kDa band upon analysis by SDS-PAGE (Fuchs et al., 1986). The 60 kDa P. stutzeri chitinase was similarly prepared. Chitinase from
Streptomyces griseus was purchased from Sigma and purified by chitin adsorption and elution (Mdano et al., 1977).
